Introduction
Genomic instability, telomere wear, epigenetic alterations, mitochondrial dysfunction, loss of proteostasis, dysregulation of nutrient sensing, cellular senescence, stem cell depletion, and altered intercellular communication were the nine original characteristics of aging proposed by López-Otín and colleagues in 2013.
In the last 10 years, several scientists have delved into research on aging, which has made it possible to formulate new characteristics of aging such as compromised autophagy, alteration of the microbiome, altered mechanical properties, splicing dysregulation and inflammation, among other emerging characteristics.
Personally, I like to turn these hallmarks of ageing on their head and talk about the hallmarks of longevity, because if we know why we age, we can look for therapeutic targets that, although they do not intend in themselves to cure ageing, they do delay its process, to achieve a healthy, happy and productive life in the elderly.
Mitochondrial Dysfunction, One of the 20 Hallmarks of Aging
At the level of mitochondrial dysfunction, there are two aspects, on the one hand the overproduction of Free Radicals and on the other the Biogenesis and Mitochondrial integrity. Under physiological conditions, ROS participate in processes such as immune response, inflammation, as well as synaptic plasticity, learning and memory.
As cells and organisms age, increased production of reactive oxygen species (ROS) result, and the efficiency of the respiratory chain tends to decrease, thus increasing electron leakage and reducing ATP generation.
During aging, a process of mitochondrial depolarization and uncoupling of OXPHOS has been found due to the opening of the mitochondrial permeability transition pore (mPTP), this prolonged opening is associated with pathological conditions.
We can talk about 10 ways to stimulate mitochondrial function in order to improve longevity.
During the conference I will also mention the key active ingredients to act.
