Longvida®

Longvida® contains the natural antioxidant curcumin, optimized for maximum bioavailability into blood and target tissues. It supplies significant levels of free (not glucuronidated or inactivated) curcumin, and this has led to clinically and statistically significant results in clinical trials.

·         Longvida® is effective, supporting nervous system function with small dosages: One small, single daily dose of Longvida® offered significant health benefits in just 30 days. An open trial on Longvida® showed 80% of people reported cognitive function improved significantly or somewhat, using low-dose Longvida®. In another human trial, a single dose of Longvida® in humans has shown the ability to bind to brain aging-related aggregates [ie amyloid-beta].

Memophenol™

Memophenol™ is a unique patent-pending solution coming from the association of selected monomers of flavonols and stilbenes from French grape and flavonols, phenolics acids and anthocyanins specific from wild blueberry.

Demonstrated synergistic association of grape and blueberry

Four blueberry active compounds, (quercetin glucuronide, chlorogenic acid, petunidin-3-glucoside and malvidin-3-glucoside) have shown, when administrated at the same dosage, to be on average three to six times more absorbed when combined with grape monomers of flavonols.

Memophenol™ is a synergistic formulation which polyphenolic compounds have been demonstrated to be highly bioavailable and distributed into brain.

Memophenol: How Does it work?

Memophenol™ has demonstrated complementary and synergistic actions on the 4 parameters involved in hippocampal-dependent memory impairment:

-       Brain plasticity: Improves markers (proteins and factors) involved in synaptic signalizations.

-       Neurogenesis: Improves the maduration of new-bornn neurons and preserves the brain cells from death.

Bacopa monnieri

Bacopa monnieri is a Nootropic herb that has been used in traditional medicine for longevity and cognitive enhancement. Bacopa monnieri has also been shown to improve specific domains of cognition, sensitive to age associated cognitive decline (particularly processing speed and memory). These Bacopa monnieri dependent improvements may be due to the increase in specific neuro-molecular mechanisms implicated in the enhancement of neural connections in the brain (i.e. synaptogenesis). In particular, a number of animal studies have shown Bacopa monnieri consumption upregulates calcium dependent kinases in the synapse and post-synaptic cell, crucial for strengthening and growing connections between neurons. These effects have been shown to occur in areas important for cognitive processes, such as the hippocampus. As Bacopa monnieri has shown neuro-molecular mechanisms that encourage synaptogenesis, while cognitive training enhances brain connectivity, Bacopa monnieri supplementation could theoretically enhance and strengthen synaptic changes acquired through cognitive training.

Improves cerebral and peripheral circulation, reduce serum cholesterol levels and act as trophic for nerve cell. Ginkgo biloba extract is a well-defined extract obtained from Ginkgo biloba leaves according to a standardized method. It has been used extensively for the treatment of diseases related to the central nervous system including neurosensory disturbances, cerebrovascular insufficiency, peripheral vascular disturbances, and degenerative dementia. The potential use of has also been suggested for the treatment of psychiatric disorders such as anxiety and depression.

Ginkgo Biloba extract has the ability to produce neuroprotection due to its chemical composition and the synergy of its components. We describe several neuroprotective mechanisms of action of such as antioxidant effects, modulation of neurotransmission, neuroendocrine regulation, and upregulation of neurotrophic factors, which underlie its potential therapeutic effect on psychiatric disorders.

Choline bitartrate

Choline is important for cell membranes and for the production of the neurotransmitter acetylcholine, which has a role in memory and muscle control. Choline bitartrate and other choline supplements are claimed to enhance athletic performance, reduce cholesterol levels, protect the liver from alcohol-related damage, lower blood pressure, control mood swings, improve memory and treat Alzheimer's disease.

Pycnogenol®

Pycnogenol® is a patent extract made exclusively from the bark of French maritime pine trees (Pinus pinaster). Pycnogenol® contains consistent proportions of bioflavonoids and is standardized to contain 70 ± 5% procyanidins. This dosage of Pycnogenol® has previously been shown to enhance memory performance after 3 months of supplementation.

Improvement of the cognitive function in adults

Reduced oxidative stress

In all three investigations with adults and baby boom­ers, the level of oxidative stress in blood was reduced by 20–30% versus inclusion. Whether there is a causal relationship between the strong anti-oxidative effect and the improvement of cognitive function remains open for discussion. It is known that oxidative stress is a critical factor in the aging process. However, there are growing evidences that suggest NO also plays a role in the aging process of the brain.

QUATREFOLIC

Quatrefolic® does not mask vitamin B12

Quatrefolic®, as (6S)-5-methyltetrahydrofolate, does not mask vitamin B12 deficiency and should represents the right way to decrease homocysteine blood concentration, diminishing the potential for masking.

Generations of folate

The terms "Folate" and "folic acid", also known as vitamin B9, are often used interchangeably to indicate a group of water soluble B-vitamins.

Actually there are differences in the forms:

1st generation - Food folate

Refers to the various tetrahydrofolate derivatives naturally present in foods.

2nd generation - Folic acid

Got its name from folium, the Latin word for leaf, when it was isolated from spinach in 1941.  It is a synthetic oxidized  molecule, that does not occur in nature but can be utilized by the human body as a precursor to form natural folates that are biologically active.

Folic acid lacks coenzyme activity and must be reduced to the metabolically active form within the cell, through a series of biochemical steps before it can be used by the body's cells in vital metabolic pathways such as DNA production, cell reproduction and homocysteine metabolism.

3rd generation - (6S)-5-methyltetrahydrofolate calcium salt 

In 1989 Bioresearch S.p.A. was the first company to evaluate the real importance of 5-methyltetrahydrofolate and filed the use of stable pharmaceutical composition of 5-methyltetrahydrofolate.

The calcium salt of 5-methyltetrahydrofolate is available commercially and represents the third generation of folate. Until now, 5-methyltetrahydrofolate calcium salt was the only folic acid derivative available on the market, and able to penetrate the body cells without the need of further metabolism process.

4th generation: Quatrefolic®

The goal for Gnosis' R&D was to develop an innovative folate salt form able to overcome the existing calcium salt form limitations related to stability and poor solubility.

In February 2008, Gnosis patented a new generation of folate derivative, brand named Quatrefolic®, (6S)-5-methyltetrahydrofolate glucosamine salt. 

Quatrefolic® represents the fourth generation folate endowed with long lasting stability as well as a peculiarly high water solubility, improved bioavailability and well established safety.

Conversion of tetrahydrofolic acid into 5-methyltetrahydrofolate is mediated by the action of the enzyme methylentetrahydrofolate reductase. In individuals with a genetic defect of the methylentetrahydrofolate reductase (MTHFR) this enzyme conversion is limited, consequently it will predispose these individuals to an increased risk for certain disease conditions.

Supplementation with 5-methyltetrahydrofolate is preferable to the one with folic acid, as it is immediately bioavailable to react with homocysteine to avoid the possibility to incur in hyperhomocysteinemia.
